SHANK3 is a multidomain scaffolding protein located at the postsynaptic density of excitatory synapses. It anchors glutamate receptors, ion channels, and signaling proteins to the actin cytoskeleton, playing a vital role in synaptic structure and function. SHANK3 is involved in receptor tyrosine kinase signaling, including the ERK-MAPK and PI3K pathways, which are essential for synaptic plasticity and neuronal survival. Mutations or deletions in SHANK3 are strongly associated with autism spectrum disorders and Phelan-McDermid syndrome, often resulting in impaired language and social communication. In neurodegenerative disease, SHANK3 dysfunction contributes to synaptic disassembly and cognitive decline.
