Metabotropic glutamate receptor 5 (mGluR5) is a G-protein-coupled receptor that modulates excitatory neurotransmission and synaptic plasticity in the central nervous system. It plays a critical role in learning, memory, and neurodevelopment. Dysregulation of mGluR5 signaling has been implicated in a range of neurodegenerative and neuropsychiatric disorders, including Alzheimer’s disease, Huntington’s disease, and fragile X syndrome. Overactivation of mGluR5 can lead to excitotoxicity, neuroinflammation, and synaptic dysfunction. Conversely, pharmacological inhibition of mGluR5 has shown neuroprotective effects in preclinical models by reducing amyloid-beta accumulation, restoring synaptic function, and attenuating cognitive deficits. As a modulator of glutamatergic signaling, mGluR5 represents a promising therapeutic target for restoring neural circuit balance and preventing neurodegenerative progression. Ongoing clinical trials are evaluating mGluR5 antagonists for their efficacy in treating cognitive and behavioral symptoms associated with neurodegeneration.
