Dicer is a ribonuclease III enzyme essential for the biogenesis of microRNAs (miRNAs), which regulate gene expression post-transcriptionally. It processes precursor miRNAs into mature ~22-nucleotide miRNAs that guide the RNA-induced silencing complex (RISC) to target mRNAs. In the nervous system, Dicer is critical for neuronal differentiation, synaptic plasticity, and stress responses. Dysregulation of Dicer or miRNA pathways has been implicated in neurodegenerative diseases, including Alzheimer’s, ALS, and Huntington’s disease. Loss of Dicer function leads to widespread transcriptomic imbalance and neuronal vulnerability, making it a key player in epigenetic regulation and a potential therapeutic target in neurodegeneration.
