Prolactin (PRL) is a neuroendocrine pituitary hormone that acts mainly on the mammary gland to stimulate lactation, but it also has pleiotropic effects in males and females. Non-glycosylated prolactin is created in the pituitary and held in storage granules before release, while glycosylated prolactin is said to be constantly secreted. Prolactin is made in the anterior pituitary in all mammal species, and its production is regulated by tonic inhibition from the hypothalamus. Prolactin is formed peripherally in humans as well. Prolactin expression is minimal in early pregnancy and rises later on. The prolactin receptor (PRLR) is a cytokine hematopoietic receptor protein. It's thought that the prolactin molecule binds to two receptor molecules. In addition to its lactogenic activity; peripherally produced prolactin plays roles in breast and prostate cancer development; regulation of reproductive function; and immunoregulation.
