LGMN/Legumain is a cysteine protease with a strict asparaginyl bond hydrolysis specificity. LGMN/Legumain may be involved in bacterial peptides and endogenous proteins' lysosomal/endosomal processing for MHC class II presentation. LGMN seem to be autocatalytic and are activated by an acidic pH. Protein expression occurs in monocytes that differentiate into dendritic cells. After lipopolysaccharide is expressed in mature dendritic cells, a fully developed, functioning enzyme is generated. Overexpression of this gene has been linked to a wide range of solid tumours. There is a pseudogene for this gene on chromosome 13. Only two alternative spliced transcript variants have been verified to be biologically valid, but numerous others have been reported. LGMN is related to two distinct diseases: schistosomiasis and Cst3-related cerebral amyloid angiopathy.
