Fumaratase catalyses the reversible hydration/dehydration of fumarate to S-malate and is involved in the tricarboxylic acid or Krebs cycle. Fumarate exists in two forms: a cytosolic form and an N-terminal extension of the mitochondrial form. The N-terminal extended form of the protein is intended for the mitochondrion, where the same process as in the cytoplasm takes place. Fumarate is considered a tumor suppressor, and its deficiency may lead to progressive encephalopathy, cerebral atrophy, and developmental delay.
