Human CSF1R/CD115 Recombinant Protein

M-CSFR encoded by the proto-oncogene c-fms is the receptor for colony stimulating factor 1 (CSF1R); a cytokine involved in the proliferation; differentiation; and activation of macrophages. The extracellular ligand-binding domain, a single membrane-spanning segment, and an intracellular tyrosine kinase domain make up this cell surface glycoprotein. The receptor kinase is activated by the binding of CSF1, resulting in autophosphorylation of receptor subunits and the phosphorylation of a number of cellular proteins on tyrosine residues. The CSF1R is a tyrosine kinase receptor that is necessary for macrophage development and has a significant role in hematopoiesis. CSF1 and its receptor (CSF1R; product of c-fms proto-oncogene) were initially implicated as essential for normal monocyte development as well as for trophoblastic implantation. The apparent function of CSF1/CSF1R in normal mammary gland development is fascinating, given that this receptor/ligand pair has also been linked to breast cancer biology where aberrant expression of CSF1 and its receptor is linked with tumor cell invasion and harmful clinical outcome. CSF1R gene expression is controlled by numerous steroid hormones (glucocorticoids and progestins) as well as several bHLH transcription factors. Tumor cell CSF-1R expression appears to be regulated by additional hormones, some of which are involved in normal lactogenic differentiation. CSF1 and CSF1R, on the other hand, have been shown to play a role in breast development throughout pregnancy and lactation. The study of CSF1 and CSF1R in normal and neoplastic mammary development might identify connections between growth factor-induced biological changes in the breast during pregnancy and tumor progression.
