C5a is a protein fragment released from complement component C5. During the classic, alternative, and lectin pathways of complement activation, C5a convertase on the C5 alpha-chain cleaves C5a to generate C5a in humans. C5a has a core region made up of four anti-parallel alpha-helices joined by three disulfide bonds, as well as a structured C-terminal tail. Carboxypeptidase B cleaves C5a to a 73-amino acid low activity form, C5a des-Arg. C5a is a highly inflammatory mediator, as well as a strong chemotactic attractant for neutrophils and other leukocytes. It increases histamine production, promotes permeability in the blood vessels, and increases the levels of several cytokines from leukocytes.
