Human Complement C5a Recombinant Protein

C5a is a protein fragment released from complement component C5. During the classic, alternative, and lectin pathways of complement activation, C5a convertase on the C5 alpha-chain cleaves C5a to generate C5a in humans. C5a has a core region made up of four anti-parallel alpha-helices joined by three disulfide bonds, as well as a structured C-terminal tail. Carboxypeptidase B cleaves C5a to a 73-amino acid low activity form, C5a des-Arg. C5a is a highly inflammatory mediator, as well as a strong chemotactic attractant for neutrophils and other leukocytes. It increases histamine production, promotes permeability in the blood vessels, and increases the levels of several cytokines from leukocytes. It promotes neutrophil-endothelial cell adhesion, enhances the humoral and cell-mediated immune response. Carboxypeptidases act on C5a, converting it to the less active C5adesArg. C5a and C5adesArg stimulate the innate immune response by acting through a classical G protein-coupled receptor, CD88. Their actions at the non-G protein-coupled receptor C5L2 (or GPR77) are still unknown. C5a has a wide range of activities since it is widely expressed throughout the body. C5a has been identified as a key pathogenic player in several immuno-inflammatory diseases, making C5a inhibition a viable therapeutic approach.
