BNIP3L is required for the formation of mitochondria during lens fiber cell remodeling, and BNIP3L is also necessary for the retention of endoplasmic reticulum and Golgi apparatus. BNIP3L localizes to the endoplasmic reticulum and Golgi apparatus of wild-type newborn mouse lenses and is contained within mitochondria, endoplasmic reticulum and Golgi apparatus isolated from adult mouse liver. BNIP3L is expressed as the cells accumulate keratin bundles, and it stimulates mitophagy to destroy the cells' last remaining 'living' feature, bringing the hair follicle from "alive" to "dead." Tissue hypoxia induces HIF1A/HIF-1 stabilization, resulting in enhanced expression of the mitophagy receptor BNIP3L/NIX. BNIP3L-dependent mitophagy results in a metabolic shift toward glycolysis essential for RGC neurogenesis. For example, the BNIP3L protein may be a viable therapeutic target for ischemic stroke.
