The G2/M transition and the ensuing mitosis are tightly orchestrated to ensure that cells divide only after DNA replication is complete and the genome is intact. The central driver of mitotic entry is the complex of Cyclin B1 with the cyclin-dependent kinase CDK1/CDC2. This complex is held inactive by inhibitory phosphorylation imposed by the kinase Wee1, and its activation requires removal of those phosphates by the phosphatase CDC25C. Once activated, the Cyclin B1-CDK1 complex phosphorylates a broad set of substrates to trigger nuclear envelope breakdown, chromosome condensation, and spindle assembly. Among the key mitotic kinases, Aurora A regulates centrosome maturation and bipolar spindle formation, Aurora B functions within the chromosomal passenger complex to govern kinetochore-microtubule attachment and the spindle assembly checkpoint, and PLK1 coordinates mitotic entry, centrosome function, and cytokinesis.
