T-complex protein 1 subunit alpha (TCP1α) is a core component of the eukaryotic chaperonin-containing TCP1 complex (CCT or TRiC), a cytosolic molecular chaperone essential for the ATP-dependent folding of key cytoskeletal proteins such as actin and tubulin. TCP1α is ubiquitously expressed and highly conserved, playing a central role in maintaining proteostasis under both physiological and stress conditions. As part of the CCT complex, TCP1α assists in the proper folding of a wide range of client proteins involved in cell structure, intracellular transport, and signal transduction. Like heat shock proteins (e.g., HSP70), TCP1α is upregulated in response to cellular stress and exhibits ATPase activity, underscoring its role in protein quality control. In the context of neurodegenerative disease, TCP1α has gained attention for its involvement in mitigating protein misfolding and aggregation—hallmarks of disorders such as Alzheimer’s, Parkinson’s, and Huntington’s disease. Impaired chaperonin function can lead to the accumulation of toxic protein species, disrupted cytoskeletal dynamics, and neuronal dysfunction. Moreover, TCP1α may influence the degradation of misfolded proteins via the ubiquitin-proteasome system and autophagy pathways. Given its critical role in neuronal proteostasis and cytoskeletal integrity, TCP1α is emerging as a potential therapeutic target for modulating protein folding and preventing neurodegeneration.
