SHANK1 is a postsynaptic density (PSD) scaffolding protein that integrates neurotransmitter receptors and signaling molecules into the cortical cytoskeleton. It plays a key role in synaptic organization, plasticity, and signal transduction at excitatory synapses. SHANK1 recruits proteins such as betaPIX and PAK to dendritic spines, regulating spine morphology and synaptic strength. It also interacts with ionotropic and metabotropic glutamate receptor complexes, contributing to synaptic signaling fidelity. Alternative splicing of SHANK1 influences its interaction network, affecting synaptic composition during development and in the adult brain. Altered SHANK1 expression or function has been linked to cognitive deficits and neuropsychiatric conditions, highlighting its relevance in neurodegenerative disease mechanisms.
