Background: | This gene is a member of the N-myc downregulated gene family which belongs to the alpha/beta hydrolase superfamily. The protein encoded by this gene is a cytoplasmic protein involved in stress responses, hormone responses, cell growth, and differentiation. The encoded protein is necessary for p53-mediated caspase activation and apoptosis. Mutations in this gene are a cause of Charcot-Marie-Tooth disease type 4D, and expression of this gene may be a prognostic indicator for several types of cancer. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. |
UniProt Protein Function: | NDRG1: a hypoxia-inducible anti-apoptotic protein. Promotes proliferation and correlates with invasion, metastasis and poor prognosis in many cancers. Is involved in growth arrest and cell differentiation during development. A ubiquitous Rab4a effector protein that modulates angiogenesis and is involved in vesicular recycling of E-cadherin. Colocalizes with transferrin during the recycling in cells. NDRG1 knockdown delays the recycling rate of transferrin, while its overexpression increases the rate of transferrin recycling. Interacts with SIRT1/p53 signaling to attenuate hypoxic injury in human trophoblasts. Its expression is induced by an elevation of free intracellular calcium ion caused by nickel ion exposure. Highly expressed in placental membranes, prostate, kidney, small intestine, ovary, peripheral nerves and Schwann cells. Defects in NDRG1 are the cause of Charcot-Marie-Tooth disease type 4D (CMT4D). |
