Mediator of DNA Damage Checkpoint 1 (MDC1) is a key regulator of the DNA damage response (DDR), orchestrating the recruitment of repair proteins to sites of double-strand breaks. In post-mitotic neurons, where genomic integrity is critical for long-term function, MDC1 plays a vital role in maintaining neuronal viability. Dysregulation of MDC1 impairs DNA repair efficiency, contributing to the accumulation of DNA lesions observed in neurodegenerative diseases such as Alzheimer’s and Huntington’s disease. MDC1 also interacts with chromatin remodeling complexes and checkpoint kinases, linking it to broader epigenetic and cell cycle control mechanisms. As DNA damage and genomic instability are emerging hallmarks of neurodegeneration, MDC1 represents a promising target for therapeutic strategies aimed at enhancing neuronal resilience and genomic maintenance.
