![]() | Immunofluorescence staining of HepG2 cells with PACO62959 at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
![]() | IHC image of PACO62959 diluted at 1:300 and staining in paraffin-embedded human liver tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. |
| Background: | Calcium-dependent phospholipase A2 that selectively hydrolyzes glycerophospholipids in the sn-2 position with a preference for arachidonoyl phospholipids. Has a much weaker activity than PLA2G4A. Isoform 3 has calcium-dependent activity against palmitoyl-arachidonyl-phosphatidylethanolamine and low level lysophospholipase activity but no activity against phosphatidylcholine. Isoform 5 does have activity against phosphatidylcholine. |
| Synonyms: | Cytosolic phospholipase A2 β (cPLA2-beta) (EC 3.1.1.4) (Phospholipase A2 group IVB), PLA2G4B |
| UniProt Protein Function: | PLA2G4B: a calcium-dependent phospholipase A2 that catalyzes the release of arachidonic acid from membrane phospholipids. Selectively hydrolyzes glycerophospholipids in the sn-2 position with a preference for arachidonoyl phospholipids. Has a much weaker activity than PLA2G4A. Not inhibited by pyrrolidine-2 and Wyeth-1. The N-terminal C2 domain associates with lipid membranes and mediates its regulation by presenting the active site to its substrate in response to elevations of cytosolic Ca2+. Four isoforms of the human protein are produced by alternative splicing. Isoform 3 translocates to membrane vesicles in a calcium-dependent fashion and has activity against palmitoyl-arachidonyl-phosphatidylethanolamine and low level lysophospholipase activity but no activity against phosphatidylcholine.Protein type: Phospholipase; EC 3.1.1.4; Lipid Metabolism - arachidonic acidChromosomal Location of Human Ortholog: 15q11.2-q21.3Cellular Component: cytosol; extracellular region; mitochondrial inner membraneMolecular Function: calcium-dependent phospholipase A2 activity; lysophospholipase activity; phospholipase A1 activity; phospholipase A2 activityBiological Process: glycerophospholipid catabolic process; phosphatidic acid biosynthetic process; phospholipid metabolic process |

| NCBI Summary: | This gene encodes a member of the cytosolic phospholipase A2 protein family. Phospholipase A2 enzymes hydrolyze the sn-2 bond of phospholipids, releasing lysophospholipids and fatty acids. This enzyme may be associated with mitochondria and early endosomes. Most tissues also express read-through transcripts from the upstream gene into this gene, some of which may encode fusion proteins combining the N-terminus of the upstream gene including its JmjC domain with the almost complete coding region of this gene, including the C2 and cytoplasmic phospholipase A2 domains.
