Legumain, also known as LGMN is a lysosomal cysteine protease which is foundin all mouse tissues, however it was mainly abundant in the kidney as well asplacenta. LGMN plays an essential role in the endosomal/lysosomal degradationsystem as the Legumain deficiency causes the accumulation of pro cathepsins B, H& L, another group of lysosomal cysteine proteases. Furthermore, overexpression of LGMN in tumors is important for invasion/metastasis.
LGMN produced in Sf9 Baculovirus cells is a single,glycosylated polypeptide chain containing 426 amino acids (18-435a.a.) andhaving a molecular mass of 48.6kDa.�(Molecular size on SDS-PAGE will appear atapproximately 40-57kDa).LGMN is expressed with an 8 amino acid His tag atC-Terminus and purified by proprietary chromatographic techniques.
UniProt Protein Function: | Has a strict specificity for hydrolysis of asparaginyl bonds. Can also cleave aspartyl bonds slowly, especially under acidic conditions. May be involved in the processing of proteins for MHC class II antigen presentation in the lysosomal/endosomal system. Required for normal lysosomal protein degradation in renal proximal tubules. Required for normal degradation of internalized EGFR. Plays a role in the regulation of cell proliferation via its role in EGFR degradation. |

NCBI Summary: | This gene encodes a member of the cysteine peptidase family C13 that plays an important role in the endosome/lysosomal degradation system. The encoded inactive preproprotein undergoes autocatalytic removal of the C-terminal inhibitory propeptide to generate the active endopeptidase that cleaves protein substrates on the C-terminal side of asparagine residues. Mice lacking the encoded protein exhibit defects in the lysosomal processing of proteins resulting in their accumulation in the lysosomes, and develop symptoms resembling hemophagocytic lymphohistiocytosis.
