Cadherin-5, also known as CDH5 is a classical cadherin which belongs tothe cadherin superfamily. The CDH5 is located in a six-cadherincluster in a region on the long arm of chromosome 16 which is involved in lossof heterozygosity events in breast as well as prostate cancer.CDH5 is a calcium-dependent cell-cell adhesion glycoproteincomposed of five extracellular cadherin repeats, a transmembrane region and ahighly conserved cytoplasmic tail. Furthermore, CDH5 functionsas a classic cadherin by passing on to cells the ability to adhere in a homophilic manner, CDH5�plays a significant role in endothelial cell biology through control ofthe cohesion as well as organization of the intercellular junctions.
CDH5 produced in Sf9 Insect cells is a single, glycosylated polypeptide chain containing 583 amino acids (25-599a.a.) and having a molecular mass of 66.2kDa. (Molecular size on SDS-PAGE will appear at approximately 70-100kDa).CDH5 is expressed with an 8 amino acid His tag at C-Terminus and purified by proprietary chromatographic techniques.
UniProt Protein Function: | CDH5: Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. This cadherin may play a important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. It associates with alpha-catenin forming a link to the cytoskeleton. Acts in concert with KRIT1 to establish and maintain correct endothelial cell polarity and vascular lumen. These effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. Required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. Interacts via cadherin 5 domain with PTPRB. Interacts with TRPC4. Interacts with KRIT1. Endothelial tissues and brain. |

The encoded preproprotein is further processed to generate a mature protein. Mice lacking the encoded protein die in utero due to vascular insufficiency, caused by increased endothelial apoptosis.
