CD55belongs to the RCA (regulators of complement activation) family characterizedby 4-30 SCRs (short consensus repeats) in their plasma-exposed regions. CD55 iswidely expressed in malignant tumors and nevertheless functions as an inhibitorof the complement system.
CD55 produced in Sf9 Baculovirus cells is a single,glycosylated polypeptide chain containing 336 amino acids (35-362 a.a.) andhaving a molecular mass of 36.8kDa (Migrates at 40-57kDa on SDS-PAGE underreducing conditions).CD55 is expressed with an 8 amino acid His tag atC-Terminus and purified by proprietary chromatographic techniques.
UniProt Protein Function: | CD55: This protein recognizes C4b and C3b fragments that condense with cell-surface hydroxyl or amino groups when nascent C4b and C3b are locally generated during C4 and c3 activation. Interaction of daf with cell-associated C4b and C3b polypeptides interferes with their ability to catalyze the conversion of C2 and factor B to enzymatically active C2a and Bb and thereby prevents the formation of C4b2a and C3bBb, the amplification convertases of the complement cascade. NCBI Summary: | This gene encodes an inhibitor of both the classical and the alternative pathways of complement activation. The encoded preproprotein undergoes post-translational processing to generate a mature polypeptide anchored to the plasma membrane via a glycosylphosphatidylinositol moiety. Erythrocytes from mice deficient in the encoded protein exhibit impaired regulation of complement activation resulting in enhanced complement deposition. Mice lacking the encoded protein exhibit enhanced susceptibility to experimentally induced myasthenia gravis. This gene is located adjacent to a closely related gene on chromosome 1.
