Human Siglec-5 are Itype (Igtype) lectins belonging to the Ig superfamily. They are defined by an N-terminal Ig-like V type domain that binds sialic acid, and a variable number of Ig-like C2 type domains. CD170 is a protein that has been designated SIGLEC5, which is expressed by monocytic or myeloid lineage cells and can be found in high quantities in peripheral blood leukocytes, the spleen, bone marrow, and at lower levels in lymph nodes, lungs, appendixes, placentas. Monocytes and neutrophils express SIGLEC5, whereas leukemic cell lines with a myelomonocytic differentiation are devoid of it. Siglec5 to 11 share a high degree of sequence similarity with CD33/Siglec3 both in their extracellular and intracellular regions. They are collectively referred to as CD33 related Siglecs. The CD33-related Siglecs' unique expression pattern in the hematopoietic system is one of their most remarkable features. CD33-related Siglecs are involved in the regulation of cellular activation within the immune system, according to this data and the fact that CD33s have two conserved immunoreceptor tyrosinebased inhibition motifs (ITIMs) in their cytoplasma tails.
