Human S100A12/CAGC Recombinant Protein

There are at least 21 distinct S100 proteins, and the protein is completely soluble in ammonium sulfate at neutral pH. Protein phosphorylation, transcription factors, cytoskeleton component movement, enzyme activities, cell development and maturation, and the inflammatory response are all affected by S100 proteins. Two EF-hand calcium-binding motifs, zinc- and copper-binding protein, are present in the S100A12 molecule. S100A12 is a disulfide-linked homodimer with an interface made up mostly of hydrophobic residues. Its proinflammatory activity involves recruitment of leukocytes, promotion of cytokine and chemokine production, and regulation of leukocyte adhesion and migration. EN-RAGE is a danger associated molecular pattern (DAMP) molecule that activates innate immune cells by binding to the receptor for advanced glycation endproducts (AGER). The activation of the MAP-kinase and NF-kappa-B signaling pathways, as well as the production of proinflammatory cytokines and cell adhesion molecules ICAM1 and VCAM1, occurs when AGER binds to AGER. It also serves as a chemoattractant for monocytes and mast cells. Furthermore, it can cause mast cell degranulation and activation, resulting in the production of chemokines, histamine and cytokines, which attract even more leukocytes to the inflamed areas. It can inhibit the activity of matrix metalloproteinases; MMP2, MMP3 and MMP9 by chelating Zn2+ from their active sites.
