Peptidyl-prolyl cis-trans isomerase Pin1 (Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1/Rotamase Pin1 and PIN1/peptidyl-prolyl cis/trans isomerase (PPIase)) is a nucleus protein. PIN1 is a peptidyl-prolyl isomerase that can change the conformation of phosphoproteins, affecting protein function and/or stability. PIN1 regulates numerous proteins essential for cell-cycle progression, and its role is thought to be that of a cellular timer. PIN1 is a PPIase that controls mitosis by interacting with NIMA and reducing its mitotic promoting activity. A protein in PIN1 displays a preference for an acidic residue at the N-terminal of the isomerized proline bond. Changes in PIN1 level can have an impact on hyperproliferative diseases such as cancer. PIN1 has been linked to several cellular cycle processes. It has been claimed that PIN1 activity is required for both normal mitotic progression and reentry into the cell cycle from quiescence. PIN1 is also overrepresented in human breast cancer, and it is a target of several oncogenic pathways. The overexpression of this gene can cause cyclin-D1 upregulation and breast epithelial cell transformation in conjunction with oncogenic pathways. PIN1 may be a promising new cancer target because it is essential for breast development. Pin1 is needed for proline-directed kinase (e.g. MAPK; CDK or GSK3) signalling and, as a result, cell proliferation (in part via control of cyclin D1 levels and stability). Recent data also implicate Pin1 as playing an important role in immune responses; at least in part by increasing the stability of cytokine mRNAs by influencing the protein complexes to which they bind.
