ORC6 is a member of the origin recognition complex (ORC) which is a highly conserved heterohexameric protein complex. ORC contacts to DNA at or near sites of initiation of DNA replication and all of its six subunits are vital for initiation of DNA replication. Additionally, ORC takes part in regulation of gene expression in response to stress. ORC6 is regularly expressed throughout the cell cycle. ORC2, ORC3, ORC4 and ORC5 form a primary complex at which point ORC6 and ORC1 assemble.
ORC6 Human Recombinant produced in E.coli is a single, non-glycosylated polypeptide chain containing 275 amino acids (1-252) and having a molecular mass of 30.5kDa.ORC6 is fused to a 23 amino acid His-tag at N-terminus & purified by proprietary chromatographic techniques.
UniProt Protein Function: | ORC6L: Component of the origin recognition complex (ORC) that binds origins of replication. DNA-binding is ATP-dependent, however specific DNA sequences that define origins of replication have not been identified so far. ORC is required to assemble the pre-replication complex necessary to initiate DNA replication. ORC is composed of six subunits. In human, ORC is cell cycle-dependent regulated: it is sequentially assembled at the exit from anaphase of mitosis and disassembled as cells enter S phase. Interacts with DBF4. Belongs to the ORC6 family. |

NCBI Summary: | The origin recognition complex (ORC) is a highly conserved six subunit protein complex essential for the initiation of the DNA replication in eukaryotic cells. Studies in yeast demonstrated that ORC binds specifically to origins of replication and serves as a platform for the assembly of additional initiation factors such as Cdc6 and Mcm proteins. The protein encoded by this gene is a subunit of the ORC complex. Gene silencing studies with small interfering RNA demonstrated that this protein plays an essential role in coordinating chromosome replication and segregation with cytokinesis.
