Napsin A Aspartic Peptidase, also known as NAPSA is a member of thepeptidase A1 family. NAPSA is involved in the processing of pneumocytesurfactant precursors. Furthermore the activation peptides of aspartic proteinasestake part as inhibitors of the active site. These peptide segments/pro-parts areconsidered essential for correct folding, targeting, as well as control of theactivation of aspartic proteinase zymogens. The pronapsin A gene is expressed mostlyin lung and kidney. In addition, NAPSA translation product is expected to be afully functional, glycosylated aspartic proteinase precursor which contains anRGD motif as well as an additional 18 residues at its C-terminus.
NAPSA Human Recombinant produced in E.Coli is a single, non-glycosylatedpolypeptide chain containing 380 amino acids (64-420 a.a) and having amolecular mass of 40.9kDa.NAPSA is fused to a 23 amino acid His-tag at N-terminus& purified by proprietary chromatographic techniques.

The encoded preproprotein is proteolytically processed to generate an activation peptide and the mature protease. The activation peptides of aspartic proteinases function as inhibitors of the protease active site. These peptide segments, or pro-parts, are deemed important for correct folding, targeting, and control of the activation of aspartic proteinase zymogens. The encoded protease may play a role in the proteolytic processing of pulmonary surfactant protein B in the lung and may function in protein catabolism in the renal proximal tubules.
