This kit uses Sandwich-CLIA as the method. The micro CLIA plate provided in this kit has been pre-coated with an antibody specific to Human MYCBP. Standards or samples are added to the appropriate micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human MYCBP and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human MYCBP, biotinylated detection antibody and Avidin-HRP conjugate will appear fluorescence. The Relative light unit (RLU) value is measured spectrophotometrically by the Chemiluminescence immunoassay analyzer. The RLU value is positively associated with the concentration of Human MYCBP. The concentration of Human MYCBP in the samples can be calculated by comparing the RLU of the samples to the standard curve.
UniProt Protein Function: | MYCBP: May control the transcriptional activity of MYC. Stimulates the activation of E box-dependent transcription by MYC. Belongs to the AMY1 family. |
UniProt Protein Details: | Protein type:Transcription, coactivator/corepressor Chromosomal Location of Human Ortholog: 1p33-p32. 2 Cellular Component: centrosome; mitochondrion; cytoplasm; nucleus Molecular Function:protein binding; transcription coactivator activity Biological Process: transcription, DNA-dependent; regulation of transcription, DNA-dependent; spermatogenesis |
NCBI Summary: | The protein encoded by this gene binds to the N-terminus of the oncogenic protein C-MYC, enhancing the ability of C-MYC to activate E box-dependent transcription. The encoded protein is normally found in the cytoplasm, but it translocates to the nucleus during S phase of the cell cycle and associates with C-MYC. This protein may be involved in spermatogenesis. This gene can be silenced by microRNA-22. Two transcript variants, one protein-coding and the other probably not protein-coding, have been found for this gene. [provided by RefSeq, Nov 2011] |

As the RLU values of the standard curve may vary according to the conditions of the actual assay performance (e. g. operator, pipetting technique, washing technique or temperature effects), the operator should establish a standard curve for each test.
