IntegrinBinding Sialoprotein (IBSP) is a monomeric noncollagenous protein which belongsto the SIBLING family of extracellular matrix (ECM) proteins. IBSP issynthesized by platelets, odontoblasts, osteocytes, osteoblasts, osteoclasts, and bone marrow stromal cells. IBSP forms an integral part of the mineralizedmatrix and also plays a role as the main mediator of the hypertrophicchondrocytes-induced angiogenesis. IBSP which binds firmly to hydroxyapatite isrelated to the severity of histological cartilage lesions.
IBSP produced in Sf9 Baculovirus cells is a single, glycosylated polypeptide chain containing 309 amino acids (17-317aa) and havinga molecular mass of 34.5kDa (Migrates at 70-100kDa on SDS-PAGE under reducingconditions).IBSP is expressed with a 8 amino acid His tag atC-Terminus and purified by proprietary chromatographic techniques.
UniProt Protein Function: | Sialoprotein-2: Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Promotes Arg-Gly-Asp-dependent cell attachment. |
UniProt Protein Details: | Protein type:Secreted, signal peptide; Secreted; Cell adhesion; Motility/polarity/chemotaxis Chromosomal Location of Human Ortholog: 4q21.1 Cellular Component: extracellular space; membrane; extracellular region Biological Process: osteoblast differentiation; extracellular matrix organization and biogenesis; biomineral formation; cell adhesion |
NCBI Summary: | The protein encoded by this gene is a major structural protein of the bone matrix. It constitutes approximately 12% of the noncollagenous proteins in human bone and is synthesized by skeletal-associated cell types, including hypertrophic chondrocytes, osteoblasts, osteocytes, and osteoclasts. The only extraskeletal site of its synthesis is the trophoblast. This protein binds to calcium and hydroxyapatite via its acidic amino acid clusters, and mediates cell attachment through an RGD sequence that recognizes the vitronectin receptor.
