B1 is a member of the HMGB family, which includes three other proteins: HMGB1, HMGB2, and HMGB3. It includes two HMG box DNA-binding domains - box A and B, which are highly negatively charged at their C termini. HMGB1 is a nuclear protein that maintains nucleosomes and permits DNA bending, which is required for gene transcription. Finally, it is shown that HMGB1 can also function as a cytokine extracellularly and regulate monocyte, T cell, dendritic cell activities in inflammatory responses.
