This kit uses Sandwich-CLIA as the method. The micro CLIA plate provided in this kit has been pre-coated with an antibody specific to Human GAP43. Standards or samples are added to the appropriate micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human GAP43 and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human GAP43, biotinylated detection antibody and Avidin-HRP conjugate will appear fluorescence. The Relative light unit (RLU) value is measured spectrophotometrically by the Chemiluminescence immunoassay analyzer. The RLU value is positively associated with the concentration of Human GAP43. The concentration of Human GAP43 in the samples can be calculated by comparing the RLU of the samples to the standard curve.
UniProt Protein Function: | GAP43: This protein is associated with nerve growth. It is a major component of the motile growth cones that form the tips of elongating axons. Plays a role in axonal and dendritic filopodia induction. Identified in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. Binds calmodulin with a greater affinity in the absence of Ca(2+) than in its presence. Belongs to the neuromodulin family. 2 isoforms of the human protein are produced by alternative splicing. |
UniProt Protein Details: | Protein type:Cell surface Chromosomal Location of Human Ortholog: 3q13. 31 Cellular Component: axon; cell junction; cytoplasm; filopodium membrane; plasma membrane; postsynaptic density Molecular Function:calmodulin binding; phosphatidylserine binding; protein binding Biological Process: axon choice point recognition; cell fate commitment; glial cell differentiation; protein kinase C activation; regulation of filopodium formation; regulation of growth; response to wounding; tissue regeneration |
NCBI Summary: | The protein encoded by this gene has been termed a 'growth' or 'plasticity' protein because it is expressed at high levels in neuronal growth cones during development and axonal regeneration. This protein is considered a crucial component of an effective regenerative response in the nervous system. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008] |

As the RLU values of the standard curve may vary according to the conditions of the actual assay performance (e. g. operator, pipetting technique, washing technique or temperature effects), the operator should establish a standard curve for each test.
