Ephrins are divided into two classes based on their structures and sequence relationships: the ephrin-A (EFNA) group and the ephrin-B (EFNB) group. Ephrin receptors, make up the most significant subfamily of receptor tyrosine kinase (RTK) family. EphB6 is an unusual Eph receptor; lacking catalytic capacity due to alterations in its kinase domain. Interestingly, increased metastatic activity is linked with a decreased EphB6 receptor level in several tumour types, including breast cancer. This highlights the potential of EphB6 as a tumour aggressiveness suppressor. EphB6 inhibits cancer invasion through c-Cbl-dependent pathways, morphological changes, and cell adhesion; these findings suggest that EphB6 might be an important prognostic indicator and a potential therapeutic target. EphB6 can both stimulate and inhibit cell adhesion and migration; a study found that tyrosine phosphorylation of the receptor by an Src family kinase serves as a mechanism for the functional shift. Furthermore, Ephrin-B2 may be a physiologically active ligand for the EphB6 receptor.
