5a is a glycoprotein that is generated enzymatically and belongs to the anaphylatoxins family of structurally and functionally related proteins. C5a convertase, an enzyme that transforms the C5 alpha-chain into C5a, creates the C5a fragment. Human C5a has four alpha-helices plus three intrachain disulfide bonds that create a triple loop structure. Human C5a is 60% and 54% identical to mouse and rat C5a, respectively. C5a binds to a signaling G-protein coupled receptor (C5aR/CD88) as well as a nonsignaling GPCR called C5L2. Cd88 activation induces neutrophil chemotaxis and endothelial cell activation. It also causes macrophages and neutrophils to release hydrogen peroxide, as well as stimulating basophiles and mast cells to secrete histamine.
