Calsequestrin-1 is an isoform of calsequestrin which are a calcium-binding protein of the sarcoplasmic reticulum. It aids in the retention of calcium in the sarcoplasmic reticulum during a muscular contraction, even though the concentration of calcium in the sarcoplasmic reticulum is considerably higher than that of the cytosol. Two forms of calsequestrin have been identified: Calsequestrin-2 and Calsequestrin-1. Calsequestrin-1 is found in fast skeletal muscle. Calsequestrin-bound calcium (through a calcium release channel) activates muscular contraction. The active protein consists of a more random coil. When calcium binds, the protein's alpha-helical content rises from 3 to 11 percent. Casein kinase 2 phosphorylates both forms of calsequestrin, but the cardiac form is phosphorylated far more rapidly and to a greater extent. Calcium ions are also absorbed by the intestines via Calsequestrin-1, which is secreted there and prevents bacteria from getting them.
