![]() | Western Blot. Positive WB detected in: Hela whole cell lysate, NIH/3T3 whole cell lysate, A549 whole cell lysate, Rat lung tissue, Rat kidney tissue, Mouse heart tissue. All lanes: ARHGAP18 antibody at 4.5µg/ml. Secondary. Goat polyclonal to rabbit IgG at 1/50000 dilution. Predicted band size: 75, 71 kDa. Observed band size: 75 kDa. |
![]() | Immunofluorescence staining of HepG2 cells with PACO56074 at 1:66, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
![]() | IHC image of PACO56074 diluted at 1:200 and staining in paraffin-embedded human small intestine tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. |
Background: | Rho GTPase activating protein that suppresses F-actin polymerization by inhibiting Rho. Rho GTPase activating proteins act by converting Rho-type GTPases to an inactive GDP-bound state. Plays a key role in tissue tension and 3D tissue shape by regulating cortical actomyosin network formation. Acts downstream of YAP1 and inhibits actin polymerization, which in turn reduces nuclear localization of YAP1. Regulates cell shape, spreading, and migration. |
Synonyms: | Rho GTPase-activating protein 18 (MacGAP) (Rho-type GTPase-activating protein 18), ARHGAP18 |
UniProt Protein Function: | ARHGAP18: a GTPase activator for the Rho-type GTPases, converting them to inactive GDP-bound states. Two alternatively-spliced isoforms have been described.Protein type: GAPs, Rac/Rho; GAPsChromosomal Location of Human Ortholog: 6q22.33Cellular Component: cytoplasm; cytosolMolecular Function: GTPase activator activityBiological Process: regulation of actin cytoskeleton organization and biogenesis; regulation of actin filament polymerization; regulation of cell shape; regulation of small GTPase mediated signal transduction; small GTPase mediated signal transduction |
