Background: | The protein encoded by this gene is a protease that functions as a deubiquitinating enzyme. The encoded protein is thought to help regulate the spindle assembly checkpoint by preventing early anaphase onset. This protein specifically deubiquitinates CDC20, which stabilizes the anaphase promoting complex/cyclosome. |
UniProt Protein Function: | USP44: Deubiquitinase that plays a key role in the spindle checkpoint by preventing premature anaphase onset. Acts by specifically mediating deubiquitination of CDC20, a negative regulator of the anaphase promoting complex/cyclosome (APC/C). Deubiquitination of CDC20 leads to stabilize the MAD2L1-CDC20- APC/C ternary complex (also named mitotic checkpoint), thereby preventing premature activation of the APC/C. Belongs to the peptidase C19 family. USP44 subfamily. |
UniProt Protein Details: | Protein type:Protease; EC 3.4.19.12; Ubiquitin-specific protease Chromosomal Location of Human Ortholog: 12q22 Cellular Component: microtubule cytoskeleton; nucleolus; nucleus Molecular Function:protein binding; ubiquitin-specific protease activity; zinc ion binding Biological Process: cell division; mitosis; proteasomal ubiquitin-dependent protein catabolic process; protein deubiquitination |
NCBI Summary: | Modification of cellular proteins by ubiquitin is an essential regulatory mechanism controlled by the coordinated action of multiple ubiquitin-conjugating and deubiquitinating enzymes. USP44 belongs to a large family of cysteine proteases that function as deubiquitinating enzymes (Quesada et al., 2004 [PubMed 14715245]).[supplied by OMIM, Mar 2008] |
