| UniProt Protein Function: | Afadin: plays a role in adhesion by participating, along with E-cadherin and catenin, in the organization of homotypic, interneuronal and heterotypic cell-cell adherens junctions (AJs). Essential for the association of nectin and E-cadherin. Contains 1 forkhead-associated (FHA) domain, a phosphopeptide recognition domain; 1 dilute (DIL) domain, a myosin-like domain which may play a functional role in cellular processes or vectorial vesicle transport; 1 PDZ domain, which frequently binds the carboxyl-terminal sequences of proteins or phosphatidylinositol 4,5-bisphosphate in the plasma membrane; and 2 RA domains, which are associated with RasGTP effector activity. Five alternatively spliced isoforms have been described. Isoform 2 does not interact with F-actin. Protein type: Oncoprotein; Motility/polarity/chemotaxis; Cell adhesion Chromosomal Location of Human Ortholog: 6q27 Cellular Component: nucleoplasm; cell-cell adherens junction; apical part of cell; cytoplasm; plasma membrane; intercellular junction; cytosol; cell junction Molecular Function: protein C-terminus binding; protein binding; Ras GTPase binding; cell adhesion molecule binding Biological Process: intercellular junction assembly and maintenance; cell-cell signaling; cell adhesion; signal transduction |

| NCBI Summary: | This gene encodes a multi-domain protein involved in signaling and organization of cell junctions during embryogenesis. It has also been identified as the fusion partner of acute lymphoblastic leukemia (ALL-1) gene, involved in acute myeloid leukemias with t(6;11)(q27;q23) translocation. Alternatively spliced transcript variants encoding different isoforms have been described for this gene, however, not all have been fully characterized.[provided by RefSeq, May 2011] |
